Lancaster was founded in 1729 by General James Lancaster as a small settlement on the Pee Dee River. By 1800, Lancaster had become a leading seaport and its economy was based on shipping and fishing. The town's population grew rapidly during the antebellum period and by 1875 it had reached 100,000 people. Lancaster experienced great prosperity during the first half of the 19th century thanks to its location at the mouth of the Santee River and its strategic position as an important transportation hub. However, after 1850s trends in industry began to decline, driving significant portions of Lancastrian society into poverty. In 1920, local officials proposed moving the city's administrative headquarters from downtown to less populated areas for fear that its central business district would be overwhelmed by growth in new businesses and services. By 1970, however, Lancaster's population had shrunk significantly due to successive waves of migration from rural areas to larger metropolitan cities throughout South Carolina. Today, little remains of Lancaster's original architecture or culture aside from a few historic buildings still standing within the city limits.

In 1814, it became a town when it was annexed by Columbia; Columbia renamed it Lancaster in honor of King George III. In 1865, Lancaster became an official state capital with its own government after being moved from Columbia to its present location following Confederate victory over Union forces during the American Civil War. The city's growth accelerated during Reconstruction,Commercial development led to heavy Poverty and crime until Hurricane Floyd struck in 1999 which destroyed most of downtown and many homes as well as causing extensive damage to buildings and infrastructure. In 2003, Lancaster experienced its first major natural disaster when severe flooding caused extensive damage to both its water supply and transportation systems which forced many people to leave their homes for weeks on end or even longer. Since that time Mayor Bobby Shaver has worked tirelessly to make amends with those who have been hurt or affected by this event while also working towards restoring essential services such as water availability and cleaning up debris from streetscape repair efforts that were necessary post-flood. In addition to his work mayor Shaver also created several city-wide initiatives such as Crowne Plaza’s “My Cityworks Program” which provides freeramentals like parks programming youth basketball games etc., storm sewer projects (including new bypasses) totaling $4 million dollars for improvements across the entire cityscape including increased safety features for residents along with better signage capabilities for pedestrians/cyclists/drivers etc.), awarded a grant worth $1 million dollars for hotel renovation/upgrades within downtown area – increase of 350 units bringing total number of units under development since inception of My CityWorks Program – marketing blitz aimed at increasing business opportunities throughout downtown area through private developers partnerships targeting primary tourist destinations such as SCFaire & beaches
