In 1785, John Dickinson and his wife, Mary, settled in the town of Nanticoke, Pennsylvania. They were the first white people to live there. The town was named for Captain Isaac Nanticoke who had been a captain in the American Revolution. Captain Nanticoke built a fort at what is now known as corner of Court Street and Market Place in 1785. He also founded the village of Nanticoke which grew up around him. In 1802, Captain Nanticoke's grandson, Isaac Newton Nanticoke Jr., moved to Nanticoke and became Mayor. He built a new courthouse in 1805 and continued to be Mayor until his death in 1829. His son-in-law, John Jordon (1768-1852), became Mayor after his father's death. John Jordon served as Mayor from 1830 to 1837 and then again from 1840 to 1848. He was also Justice of the Peace for many years. During these years, he erected many buildings in the town including the first schoolhouse (1830), jail (1835), post office (1836), United States Customhouse (1846), Methodist Church (1848), Odd Fellows Hall (1901) and an opera house/theatre called "The Grove" which opened on May 7th, 1859 by Henery Wells Davenport (). In 1861, Governor Andrew Johnson appointed Judge John Fenton as United States Marshal for Westmoreland County because local officials could not keep up with court cases that were being prosecuted by federal prosecutors based out of Philadelphia (). Judge Fenton stayed with Westmoreland County until 1865 when he resigned because he was nominated to become Chief Justice of Pennsylvania (). He is buried at Green Lawn Cemetery near Pittsburgh where his tombstone reads “ Honored Member Of The Bar And Peoples’ Political Friend” ().

The community was named after the Dutch settlement of Nantes on the Delaware River. It is located in rural Lancaster County on the east bank of the Susquehanna River. In 1764, a group of British soldiers under General Edward Braddock reached Nanticoke from Fort Duquesne and captured it from its Native American inhabitants. As part of Braddock's army, Major John Forbes built a fort here in 1768 during the Indian War with France. Forbes renamed his fort Fort Pitt (later Pittsburg). Nanticoke developed as a small farming village founded by farmers from Lancaster County and Cumberland County in 1810-15. A sawmill was started here in 1825, but it failed and was abandoned soon afterward. One of the few remaining businesses in town began operating as an insurance agency in 1859. After that time, most residents engaged in agriculture or fishing until electric transportation became available and made town development possible beginning around 1915 As with other towns throughout Lancaster County during this period, many people left Nanticoke for better opportunities during World War II fear of Japanese attack; however, no Japanese ever arrived at Nanticoke mainland nor did any attacks take place there . Although now considered an agricultural center again due to demand from nearby farms for goods exported to Japan , much remains unchanged about traditional values among locals such as closed-door dances honoring elders called " kumquatucket ," spinning wheel days on Saturday mornings before services at First Presbyterian Church , Sunday morning early wake up call called "grapefruit" because so many people gather around their kitchen table to drink grape juice (added after World War II when industry moved away), Sabbath morning prayer meeting at Seventh Day Adventist church (now known as Rite Aid) every alternate Saturday morning since 1948 ; one can still find hand grenades hidden inside well heads near home
