The present Main Street Business District was built around 1920 when two churches were merged into one: First Baptist Church and Second Presbyterian Church of Athens-Clarke County united to build a new district. In 1907, Centerville voters passed an ordinance setting up government within the limits of the original town site that included annexed areas of Holmesboro (now part of Dayton), New Albany (now part of Middletown), Clay Township (now part of Lima), and parts of Clinton Township and Uniontown Township (now part of Delaware).
Centerville was founded in 1811 by Daniel Nice and his wife Deborah. The town was originally named after the creek that ran through it. It is located on the Branch Canal, which connected the Ohio River with Lake Erie. The first settler in Centerville was John Goodloe.
