The town was founded by white settlers in 1847 and named for General Andrew Johnson, who had been president when it was founded. Tupelo's first post office opened in 1850. In 1852, the Union Pacific Railroad built a line from Meridian to Tupelo, which made the town more accessible to people living in other parts of the state. The railroad also helped to make Tupelo a commercial center and its economy boomed during its heyday. In the early 1960s, cotton production ended and jobs left Tupelo; however, it has regained some of its economic importance as an agricultural community with many businesses involved in agriculture or related industries. The town has experienced several African American majorities through gradual integration since then; however, there are still some cultural barriers that prevent Latinos and Native Americans from fully participating in local life.

The town had a population of 1,500 as of the 2010 census. It is the county seat of Jackson County. Tupelo is located on the banks of theMississippi River and its downtown area is home to many local businesses and government buildings dating back to the 1830s. The first white settler in Tupelo was David Wilkinson, who arrived in 1819 from Virginia with his family and land grant from President James K. Polk. Wilkinson's wife Mary died soon after they arrival in what would become Tupelo; Wilkinson took his son with him to live with his sister before moving out himself in 1820. By 1824, several other settlers had moved into the village including John Putnam, who built a gristmill on Dry Creek; George Stephenson also visited during this time and donated land for a schoolhouse which he named "Prentiss Academy". In 1830, Methodist Reverend Drury erected a log church at what would become First Baptist Church across from creek which served as both community center and parishioner meeting place until 1865 when it burned down; later that year it was replaced by an opera house designed by architect Elijah Davenport which still stands today within block of downtown known as Davenport Square (named for him). Other churches established during this time include Second Baptist Church (1834), Third Presbyterian Church (1835), Fourth Baptist Church (1836), African Methodist Episcopal Zion Chapel (1840-41) - all located within 5 blocks east of whereFirst Baptist now sits; Union Missionary Baptist Churches were founded adjacent to these denominations shortly thereafter but did not establish congregations near Dupont Avenue or much else outside of their own cemetery until after World War II when Interstate 10 destroyed most rural Mississippi communities
The town of Tupelo, Mississippi was founded in 1819 by a group of settlers from Kentucky. The town was named for General James Iredell, who died in 1818 while fighting in the Mexican-American War. Tupelo became a stop on the Underground Railroad during the 1850s. During the Civil War, Tupelo was attacked by Confederate forces and burned down.
