The town was founded by planters in 1881 as a cotton plantation community on the edge of Yazoo Delta. When slavery ended in 1865, Starkville became an agricultural center with large plantations and small villages. Heavily dependent on slave labor for its economic success, Starkville declined in popularity after Reconstruction until it regained some notoriety as a resort area during the late 1960s and early 1970s when white flight increased oil production in the delta region. Today, Starkville is a rural community which has experienced significant growth due to its location within easy commuting distance of both Jackson and Gulfport metropolitan areas. Its primary industry is agriculture but there are also several businesses such as a lumberyard and restaurant that have taken advantage of new technology and digital services to expand their operations. Starkville is one of the oldest towns in Mississippi and has been mentioned as early as 1792. The first settler in what is now Starkville was James Fenton who arrived in 1792 from Virginia. In 1827, Isaac Beall founded the town on land he had acquired from General Andrew Jackson. The post office was established in 1829, and became a stop on the Santa Fe Railroad. In 1893, Jesse Laughlin purchased land east of town and built a large cotton gin and mills there which remain to this day known as Laughlin Cotton Mills LLC (formerly JSCM). In 1924, Drury School opened for grades Kindergarten through Eighth Grade in an old two-story frame schoolhouse located on Elm Street near present day US Highway 83. At that time it was one of only three schools open for black students in Hinds County; all other schools were white institutions until integration began in 1965. Drury School closed during World War II due to lack of enrollment but reopened after the war with special programs for African American children called "Hinds County Negroes". The school's last student graduated from Drury School on May 10th 1957 with distinction honors! Mrs Jane Addams attended Drury School also before she founded Hull House educational camp nearby which continues to provide educational opportunities today for inner city youth ages 9-18 years old

Starkville was founded by General John Stark on November 25, 1819 as a small town in present-day Hinds County. The first post office in Starkville was established on December 5, 1820. The name "Starkville" originated from General John Stark's father-in-law Colonel James Jernigan's purchase of land in what is today Starkville for $2 per acre (0.93 km²).
