In 1892, the Mississippi Valley Railroad began construction of a line from Jackson, Mississippi to Picayune, Mississippi. The line was completed in 1903 and became part of the Gulf Coast Railway. The company merged with the Memphis and Nashville Railway in 1924 and went bankrupt in 1930. In 1935, the railroad was acquired by the Union Pacific Railroad. The company changed its name to Pan American World Airways in 1941 and ceased operations in 1945. In 1948, Pan American World Airways (PANA) leased three tankers from John F Kennedy International Airport for transportation of oil products from Texas to Miami Beach, Florida. PANA used these vessels for service between Miami Beach and Key West until 1957 when they were sold to Panamanian shipping interests. The last use of Pan American World Airways' tankers came about twelve years later when it was replaced by a new ferry service between Jacksonville Landing on Florida's east coast and Fernandina Beach on Panama's west coast which operated until 1984.

In 1871, the town of Picayune was platted by John S. Ellis and subscribed to the Jackson County Agricultural and Mechanical District. The first post office in Picayune was established on October 7, 1871. By 1880, Picayune had a population of 233 people. In 1907, the Mississippi River crossed over into Picayune from Louisiana and changed its course so that it became known as "The Big Black River". The name stuck for a few decades until it was changed back to "Picayune" in 1955. In 1861, Abraham Lincoln delivered a speech in which he called for states to unite to repel Confederate invaders. On November 12th of that year, three days after his speech, Union soldiers burned down the courthouse in Picayune while supplies were being stored inside.
