The community was named for General James Horn (1785-1851), who fought in the Mexican–American War and led U.S. forces against Mexico City's Mexicans under Santa Anna. The first house on the site of present-day Horn Lake was built by Samuel Hargrave in 1819. A post office called Horn Lake was established in 1824, and remained in operation until it was discontinued in 1922. Horn Lake became a town in 1903 when it merged with neighboring Laddsboro Township. The name "Horn" came from General James Horn'sMilitary Expedition to Texas which ended with his death there on April 21, 1841; his body lay undiscovered for several weeks because of all the rains that had fallen during that time period. In 1913, an alluvial fill created by creek flooding turned into a lake when Hurricane Hazel pushed water over a levee adjacent to what is now Main Street near where Walnut Creek empties into the lake. Asphalt roads were added to connect different parts of town during this time (later replaced with gravel). It reached its peak development around 1930 as more people started moving into the area because of good soil quality and opportunity for farming after construction of various railways brought new settlers closer to markets and jobs outside of Jackson (the city proper formed only about 10 years later). In recent decades there has been declines due to factors such as economic decline and population growth withinJackson itself; however some aspects like retail storefronts still remain open along Main Street despite not being used much anymore due to increased competition from other businesses downtown including larger stores opening up next door since Highway 65 bypasses town limits).

The community developed around an open-air cotton gin and gristmill on the banks of the Arkoma River. The mill was built in 1868 by John W. Horn and operated until 1935. The community changed hands several times before it came to rest within the borders of Horn Lake Township in 1960. The township had a population of just over 100 people when it merged with neighboring Carlton Township to form Corinth Township.

The first settlement on the present-day site of Horn Lake was made by French traders in 1714. In 1819, the United States acquired 1/4 of all of Louisiana from France and decided to give it to Mississippi as part of that acquisition. The original town on Horn Lake was called Hernando, after General Hernando de Soto, one of US president James K. Polk’s predecessors in office during the Mexican-American War (1846-1847). After Removal Act of 1830 came into effect which transferred all Indian land east of Arkansas River to the US government, Hernando became known as Greenwood County seat. After being renamed for Mississippi Governor John C. Calhoun following his election in 1849, it reverted back to its original name following his death in 1865.
