The area that is now Nicholasville was originally called "New Shawneetown" after Shawnee Chief Oconee who lived there for many years. In 1875–76 local farmers organized a grain elevator to supply Lexington with corn during an embargo against Mexico caused by US president Ulysses S. Grant's Proclamation 9g which banned all exports from Mexico except those essential to military purposes; this led to increased trade through Nicholsville between Lexington and Nicholasvill

Nicholasville has experienced many changes over its history, including becoming an industrial center during the 1800s; experiencing a tremendous growth period during World War II; becoming a bedroom community during the late 1960s and early 1970s; experiencing a decline in population during the 2000s; and finally reverting back to its rural roots after declining manufacturing jobs left the town in recent years. However, even with these changes, there has never been any indication that Nicholasville would eventually lose its identity or charm as one of Kentucky's most important small towns.

Nicholasville was founded by Nicholas Warfield in 1803. The town was named for General JohnNicholson, who fought in the American Revolution. Nicholson also donated land to the town. In 1824, Nicholasville had a school and a church. In 1827, the town had a post office and a sawmill. In 1830, the first store opened in Nicholasville. In 1835, the first Trinity Episcopal Church was built in Nicholasville. The town had four stores by 1840 and twelve homes by 1850. By 1860, there were twenty-one businesses in Nicholasville and ninety-six people living there. The population grew to one thousand by 1870 but declined to six hundred by 1880.
