The community rests on the Mississippi River and became known for its white cotton plantations until the early 20th century when those plantations were replaced by black cotton plantations. The mills closed in the 1930s but have been revived as a tourist attraction with some of the buildings still standing. Millbrook has an annual corn festival that features several traditional events such as rodeo, football games, and pie contests among other things.

Millbrook was founded in 1892 and named for Dr. James M. Millbrook, a local physician and businessman who donated land to the town. The first school in Millbrook was established in 1892 by Mrs. Rufus Crouse and Mr. Luther Witherspoon. In 1896, the first post office opened in Millbrook and was called "Newbern". In 1903, the millinery business of Fannie Beaman & Son moved to Millbrook from Mobile and continued to operate until 1951 when it closed its doors for good after manufacturing silk sashes for many years. In 1912, Drs John Aiken & Edward Hadden purchased land east of what is now the mill property from James Mmillbrook and laid out a village on the site which they named "Anniston". The population of Anniston reached 100 by 1920 but declined rapidly due to declining agricultural production around Anniston Lake. By 1930 only 10 people lived there; all were employed at mills or other businesses in nearby towns such as Decatur or Mobile Bayou communities which became more populous during World War II due to munitions production around Birmingham AL and related manufacturing jobs throughout Southeast Alabama following the Pensacola Movement led by General Robert E Lee that began on April 4th 1861 with an attempt by Confederate general Jeff Davis to secede from Confederate America
