The town was founded by James Waddell and his brother John on October 10, 1853, as a depot on the Yazoo River for the delivery of goods to Mobile Bay. In 1865, Enterprise became known as "the most important cotton center in Alabama." As cotton production increased throughout the Deep South during the antebellum period, Enterprise grew into a major economic engine for the state. In 1868 it became an incorporated city with a population of 12,000 people. In 1870 Enterprise passed from corporate ownership to municipal control under the leadership of Mayor Alfred Noland and City Councilman A.J. Hargrave; this move signaled a turning point in city government in Alabama which saw increasing public participation and accountability within local governments. During these years, many new businesses were established within the limits of Enterprise including J & L Coal Company (founded 1871), Copley's Meat Market (1872), Treadway's Cotton Mill (1873), Grosse Pointe Park Railroad Depot (1875) and Rogers' Drug Store (1876). This growth led to increased demand for streetlights and other civic services which contributed to overall financial stability and prosperity until annexation by Mobile in 1901 brought about fundamental changes in local government that undermined community solidarity and commerce
In 1819, the first white settler in Enterprise was James T. McCoy who purchased land from the native tribe. The first store in Enterprise was built by John Smith and operated out of a small cabin on the banks of the Mobile River. The town grew rapidly due to its location and rich agricultural potential. In 1830, Enterprise became one of the five counties in Alabama organized by William Hance. By 1840, Enterprise had a population of 5,000 people and boasted a series of impressive public buildings including the courthouse and jail. In 1851, Enterprise became one of six states admitted to the Union as Alabama. The following year saw an escalation in violence between local farmers and settlers over access to land, which culminated with an attack on the town by some 300 Bloody Mountain men led by Elijah Blackwood that left 10 residents dead and 50+ injuries.
